Background information: The neurotoxin 1-methyl-4-phenyl pyridinium (MPP+) enters neurons through DAT (Dopamine transporter) causing specific dopaminergic neuronal apoptosis and death, providing a useful model to study Parkinson’s disease in vitro. MPP+ induces apoptosis in SH-SY5Y cells. SB-203580 is able to inhibit MPP+-induced apoptosis
Cells: SH-SY5Y, human neuroblastoma.
Damage induction: 3 mM MPP+ for 24h.
Readout: Apoptosis.
Positive control: SB-203580.
Compound treatment: Compounds are added to cells 24h before the addition of MPP+.
Turnaround time: 1 month.
